One of two culprits:
1 - the back-up bypass valve is dysfunctional
2 - the electrical feed from the backup lights circuit to the bypass valve is dead

The valve bypasses the brake fluid from the line to the calipers back into the reservoir allowing the master cylinder to compress without actuating the brakes. So, either it is not working (bound up or corroded) or the electrical circuit is not complete to actuate it. Make sure the WHITE ground wire is making good connection, don't rely on just the hitch to make that connection.

im gonna assume u have disc brakes with an anti reverse "backup"  solenoid. most feeds for the anit reverse solenoids are picked up from the backup light circuit on the tow vehicle and are fused by the same  however later  vehicles have a separate circuit fed into the towing plug if factory equiped   look in the fuse box and make sure the towing reverse fuse is in the slot or not blown
However if u have drum brakes the lockout is totally mechanical   a small pin inserted into the coupler locks out the plunger action  or on top the knob disables it
